Canola 2-Dimensional Proteome Profiles under Osmotic Stress and Inoculation with Pseudomonas Fluorescens FY32

Abstract
In order to use the potential of plant growth-promoting bacteria to ameliorate the adverse effects of extreme conditions, identification of responsive proteins involved in the plant-bacteria interaction under stressful conditions could be very useful. In this aim, Sarigol (a sensitive cultivar of canola) was inoculated with P. fluorescence FY32 and undergone medium (70% FC) and extreme (50% FC) osmotic stress using polyethylene glycol. Protein profile analysis performed using 2D-PAGE technique. The results of morphological analysis indicated a decreased total dry and fresh weight, shoot and root dry weight, and shoot height under both of osmotic stress levels than control plants by comparison. Inoculated plants in comparison with non-inoculated plants had better morphological traits under normal and osmotic conditions. Protein profile analysis using Melanie 7 software introduced 21 of 161 reproducible spots with significantly changed expression level (p <0.01). These proteins were categorized into eight functional groups, including energy metabolisms, photosynthesis, channel proteins, cell-defense proteins, cell structure, gene regulation, and protein structure. The metabolism/energy-related proteins, ACC oxidase 4, and PDHB were affected by bacteria under 50% FC osmotic stress.
