Title Study of the effect of the pyrimidine compound on human serum albumin fibril formation
Type of Research Presentation
Keywords Human serum albumin, Amyloids,Pyrimidine compound, Fibrillation
Abstract Amyloid fibrils are aggregates of misfolded proteins with a beta-sheet secondary structure. They have been identified as the primary cause of several neurodegenerative disorders, including Alzheimer's disease.Because of the high prevalence of neurodegenerative diseases worldwide, the inhibition or reduction of protein fibril formation is the main goal ofthe researches. In current study, the effect of a synthetic pyrimidine compound has been examined on human serum albumin (HSA) which is a common model for protein fibrillation. Pyrimidine compounds have attracted a lot of attention due to their diverse biological activitiessuch as anti cancer, anti inflammatory and anti bacterial properties. In this research, HSA fibrillation was detected by fluorescence spectroscopy andfibrils were visualized using AFM method. First of all, HSA was treated under harsh environmentalcondition (high temperature and salt presence) to form fibrils. Then, fibrillation was examined in the presence of different concentrations of the drug. Data confirmed that our synthetic pyrimidine compound has the ability to inhibit HSA fibrillation in a concentration dependent manner. Based on our experimental results, this pyrimidine compound is suggested as a candidate for the inhibition or reduction of HSA and other proteins fibril formation. However more researches are needed to confirm the general anti fibrillation activity of this compound.
