Combined antimicrobial activity of Mentha logifolia essential oils and a bacteriophage against nosocomial isolates of Staphylococcus aureus

چکیده
Staphylococcus aureus as an opportunistic pathogen can cause a lots of infections. It is a public health problem due to increasing rate of antibiotic resistance.Combined application of natural compounds might provide an enhancing effect for control pathogenic microorganisms. So the effect of Mentha logifolia Essential oils (EOs), and an environmental isolated phages admixture to control S. aureus was investigated in this study. Materials and Methods: A total 30 Staphylococcus aureus isolates was collected from Emam Hossein Hospital, Hashtroud,Iran. One phage isolated from environmental samples of a hospital using staphyloccocus aureus (PTCC 1112) as a host and soft agar overlay method. Sensivity of each collected strain to isolated phage was evaluated by using 0.1 MOI of bacteriphage. Growth inhibition assays (GIA) of bacteriophage senstive S. aureus strains were monitored by mixing strains with different concentrations of M.logifolia EOs over a 24-h period at 37 ºC. Combined effect of EOs and phage were tested by simultaneous use of them. The results were compared with negative control of bacteria culture without EOs and phage. Result : Isolated phage showed lytic activity against 60% of S. aureus strains. GIAs results indicated that higher concentration of EOs (50 mg/ml) significantly reduced (P <0.005) all strains growth over a 48-h period when compared to the control. Results from the combined effect indicated that this combination had significantly higher inhibitory effect on tested strains than EO alone (P < 0.05). Conclusion : Accordingly this combination had synergistic effects and can be used as a potential antimicrobial against S. aureus.
