The recent development of microextraction techniques based on deep eutectic solvents (DESs) holds great promise in
developing green analytical chemistry. The use of chromatographic instruments in drug analysis is a powerful analytical
approach, but it is not practical for on-site analysis in many cases and needs sophisticated users. In this paper, we present a
spectrophotometer-based ultrasound-assisted emulsification–microextraction with a hydrophobic deep eutectic solvent for
preconcentration and quick determination of concentrations of the anticancer drug, doxorubicin. The spectrophotometer uses
96-well Corning microplates with a microliter volume that is entirely compatible with the microextraction technique. The
DES was prepared by mixing choline chloride and phenyl ethanol in a mole ratio of 1:4. The effect of different parameters
(pH, DES volume, salt concentration, and sonication time) was optimized by a multivariate approach, central composite
design, combined with desirability function. The optimized method showed excellent ability for extraction of doxorubicin
from urine samples. The method showed a wide linear range (0.3–15 μg/mL) with a low limit
