Hypertension is a common medical and social problem leading to cardiovascular diseases
worldwide. Calcium channel blockers (CCBs), especially 1,4-dihydropyridines (1,4-DHPs),
are nowadays the most used drugs in the treatment of hypertension. Nifedipine, Adalat®, as a
CCB drug is one of the prominent compounds of the family of 1,4-dihydropyridines and
belong the first generation. In subsequent generations, new derivatives of nifedipine that are
clearly more lipophilic deserve special attention. The studying the relationship between the
lipophilicity of these compounds and their mode of action has proven that the higher
generations of DHP with more lipophilicity are much more powerful drugs.
Adamantane, the smallest diamondoid, due to having unique properties such as
lipophilicity, physical and chemical stability, size, rigidity, biocompatibility, and the nontoxicity
of substances resulting from its degradation in the body, can have many applications,
including drug design and drug delivery. Therefore, modifications with adamantane as a
"lipophilic bullet" are of interest to increase the lipophilicity and stability of drugs, thereby
improving pharmacokinetics without increasing their toxicity.
