Investigating the association of rs920778T > C polymorphism in HOTAIR gene in breast cancer patients in the northwestern of Iran

Breast cancer is the most common malignancy in women worldwide, growing 3.1% worldwide each year becoming a global health problem. HOX transcript antisense RNA (HOTAIR) gene is one of the long noncoding RNAs that is involved in various processes such as epigenetic regulation of some genes, cancer cell proliferation, progression and metastasis and has been widely studied in recent years. The effect of HOTAIR polymorphism on various diseases has been shown. The present study examined the polymorphism rs920788T > C in the HOTAIR gene in women with breast cancer in the northwestern of Iran. In this case-control study, 203 women with breast cancer and 202 healthy women as controls were examined. RFLP-PCR was used to determine the genotypes. In this study, we used in silico databases to investigate the binding affinity of transcription factors, to determine the regulatory sequence, and to investigate the spatial structure change as a result of changes in the polymorphism rs920778T > C. Findings showed that TT homozygous (P-value = .00) and TC heterozygous (P-value = .000) were associated with breast cancer, while CC homozygous (P-value = .074) did not show association with breast cancer. Also, the results did not show a significant relationship with physiopathological properties. Finally, the results of this study showed the T-allele as a risk factor in patients with breast cancer in the northwestern of Iran. Also, this polymorphism can be used as a prognostic marker in breast cancer. However, confirmation of these results requires a study of this polymorphism in larger populations and other populations.
